Acid resistance of methanogenic bacteria in a two-stage anaerobic process treating high concentration

摘要: In this study, the two-stage upflow anaerobic sludge blanket (UASB) system and batch experiments were employed to evaluate the performance of anaerobic digestion for the treatment of high concentration methanol wastewater. The acid resistance of granular sludge and methanogenic bacteria and their metabolizing activity were investigated. The results show that the pH of the first UASB changed from 4.9 to 5.8 and 5.5 to 6.2 for the second reactor. Apparently, these were not the advisable pH levels that common methanogenic bacteria could accept. The methanogenic bacteria of the system, viz. Methanosarcina barkeri, had some acid resistance and could still degrade methanol at pH 5.0. If the methanogenic bacteria were trained further, their acid resistance would be improved somewhat. Granular sludge of the system could protect the methanogenic bacteria within its body against the impact of the acidic environment and make them degrade methanol at pH 4.5. The performance of granular sludge was attributed to its structure, bacteria species, and the distribution of bacterium inside the granule.

Anaerobic treatment of wastewater containing methanol in up-flow anaerobic sludge bed (UASB) reactor

摘要: The direct conversion of methanol into methane is the main process in anaerobic treatment of methanol containing wastewater. However, acetic acid can also be produced from methanol theoretically, which may probably result in an abrupt pH drop and deteriorate the anaerobic process. Therefore, it is interesting to know what would really happen in an anaerobic reactor treating methanol wastewater. In this study, an up-flow anaerobic sludge bed (UASB) reactor treating methanol wastewater was operated. The chemical oxygen demand (COD), acetic acid and pH of the effluent were monitored at different loadings and influent alkalinity. The results showed that the anaerobic reactor could be operated steadily at as low as 119 mg/L of influent alkalinity and high organic loading rate with no obvious pH drops. Volatile fatty acids accumulation was not observed even at strong shock loadings. The microorganisms in the sludge at the end of the test became homogeneous in morphology, which were mainly spherical or spheroidal in shape.

Use of gamma-irradiation pretreatment for enhancement of anaerobic digestibility of sewage sludge

摘要: The effects of ?-irradiation pretreatment on anaerobic digestibility of sewage sludge was investigated in this paper. Parameters like solid components, soluble components, and biogas production of anaerobic digestion experiment for sewage sludge were measured. The values of these parameters were compared before and after ?-irradiation pretreatment. Total solid (TS), volatile solid (VS), suspended solid (SS), volatile suspended solid (VSS), and average floc size of samples decreased after ?-irradiation treatment. Besides, floc size distribution of sewage sludge shifted from 80–100 ?m to 0–40 ?m after ?-irradiation treatment at the doses from 0 to 30 kGy, which indicated the disintegration of sewage sludge. Moreover, microbe cells of sewage sludge were ruptured by ?-irradiation treatment, which resulted in the release of cytoplasm and increase of soluble chemical oxygen demand (SCOD). Both sludge disintegration and microbe cells rupture enhanced the subsequent anaerobic digestion process, which was demonstrated by the increase of accumulated biogas production. Compared with digesters fed with none irradiated sludge, the accumulated biogas production increased 44, 98, and 178 mL for digesters fed sludge irradiated at 2.48, 6.51, and 11.24 kGy, respectively. The results indicated that ?-irradiation pretreatment could effectively enhance anaerobic digestibility of sewage sludge, and correspondingly, could accelerate hydrolysis process, shorten sludge retention time of sludge anaerobic digestion process.

Kinetic analysis of anaerobic phosphorus release during biological phosphorus removal process

摘要: Enhanced biological phosphorus removal (EBPR) is a commonly used and sustainable method for phosphorus removal from wastewater. Poly-β-hydroxybutyrate (PHB), polyphosphate, and glycogen are three kinds of intracellular storage polymers in phosphorus accumulation organisms. The variation of these polymers under different conditions has an apparent influence on anaerobic phosphorus release, which is very important for controlling the performance of EBPR. To obtain the mechanism and kinetic character of anaerobic phosphorus release, a series of batch experiments were performed using the excessively aerated sludge from the aerobic unit of the biological phosphorus removal system in this study. The results showed that the volatile suspended solid (VSS) had an increasing trend, while the mixed liquid suspended sludge (MLSS) and ashes were reduced during the anaerobic phosphorus release process. The interruption of anaerobic HAc-uptake and phosphorus-release occurs when the glycogen in the phosphorus-accumulating-organisms is exhausted. Under the condition of lower initial HAc-COD, HAc became the limiting factor after some time for anaerobic HAc uptake. Under the condition of higher initial HAc-COD, HAc uptake was stopped because of the depletion of glycogen in the microorganisms. The mean ratio of ΔρP/Δρ, Δρ/ΔρPHB, ΔρP/ΔCOD, and ΔρPHB/ΔCOD was 0.48, 0.50, 0.44, and 0.92, respectively, which was nearly the same as the theoretical value. The calibrated kinetic parameters of the HAc-uptake and phosphorus-release model were evaluated as follows: Q was 164 mg/(g °h), Q was 69.9 mg/(g °h), K was 0.005, and KCOD was 3 mg/L. An apparently linear correlation was observed between the ratio of ΔρP/ΔCOD and pH of the solution, and the equation between them was obtained in this study.

Effect of short-term atrazine addition on the performance of an anaerobic/anoxic/oxic process

摘要: In this study, an anaerobic/anoxic/oxic (AO) wastewater treatment process was implemented to treat domestic wastewater with short-term atrazine addition. The results provided an evaluation on the effects of an accidental pollution on the operation of a wastewater treatment plant (WWTP) in relation to Chemical Oxygen Demand (COD) and biological nutrient removal. Domestic wastewater with atrazine addition in 3 continuous days was treated when steady biological nutrient removal was achieved in the AO process. The concentrations of atrazine were 15, 10, and 5mg·L on days 1, 2 and 3, respectively. The results showed that atrazine addition did not affect the removal of COD. The specific NH NO reduction rate decreased slightly due to the short-term atrazine addition. However, it did not affect the nitrogen removal due to the high nitrification and denitrification capacity of the system. Total nitrogen (TN) removal was steady, and more than 70% was removed during the period studied. The phosphorus removal rate was not affected by the short-term addition of atrazine under the applied experimental conditions. However, more poly-hydroxy-alkanoate (PHA) was generated and utilized during atrazine addition. The results of the oxygen uptake rate (OUR) showed that the respiration of nitrifiers decreased significantly, while the activity of carbon utilizers had no obvious change with the atrazine addition. Atrazine was not removed with the AO process, even via absorption by the activated sludge in the process of the short-term addition of atrazine.

Process stability and microbial community composition in pig manure and food waste anaerobic co-digesters

摘要: This study assessed the effects of reducing hydraulic retention times (HRTs) from 21 days to 10.5 days when anaerobically co-digesting pig manure and food waste. Continuously stirred tank reactors of 3.75 L working volume were operated in triplicate at 42°C. Digester HRT was progressively decreased from 21 to 15 days to 10.5 days, with an associated increase in organic loading rate (OLR) from 3.1 kg volatile solids (VS)·m ·day to 5.1 kg VS·m ·day to 7.25 kg VS·m ·day . Reducing HRT from 21 days to 15 days caused a decrease in specific methane yields and VS removal rates. Operation at a HRT of 10.5 days initially resulted in the accumulation of isobutyric acid in each reactor. High throughput 16S rRNA gene sequencing revealed that this increase coincided with a shift in acidogenic bacterial populations, which most likely resulted in the increased isobutyric acid concentrations. This may in turn have caused the increase in relative abundance of bacteria, which syntrophically degrade non-acetate volatile fatty acids (VFAs) into H and CO . This, along with the increase in abundance of other syntrophic VFA oxidizers, such as suggests that VFA oxidation plays a role in digester operation at low HRTs. Reducing the HRT to below 21 days compromised the ability of the anaerobic digestion system to reduce enteric indicator organism counts below regulatory limits.

Dissolved methane in anaerobic effluent: Emission or recovery?

摘要: Various anaerobic processes have been explored for the energy-efficient treatment of municipal wastewater. However, dissolved methane in anaerobic effluent appears to be a barrier towards the energy and carbon neutrality of wastewater treatment. Although several dissolved methane recovery methods have been developed, their engineering feasibility and economic viability have not yet been assessed in a holistic manner. In this perspective, we thus intend to offer additional insights into the cost-benefit of dissolved methane recovery against its emission.

Membrane fouling control by ultrasound in an anaerobic membrane bioreactor

摘要: In this study, ultrasound was used to control the membrane fouling online in an anaerobic membrane bioreactor (AMBR). Short-term running experiments were carried out under different operating conditions to explore feasible ultrasonic parameters. The experimental results indicated that when the crossflow velocity was more than 1.0 m/s, membrane fouling could be controlled effectively only by hydrodynamic methods without ultrasound. When ultrasound was applied, an ultrasonic power range of 60–150 W was suitable for the membrane fouling control in the experimental system. The experimental results showed that the membrane fouling was controlled so well that membrane filtration resistance (Σ) could stay at 5 × 10 m for more than a week with the crossflow velocity of 0.75 m/s, which equaled the effect of crossflow velocity of more than 1.0 m/s without ultrasound.

Short-term effects of excessive anaerobic reaction time on anaerobic metabolism of denitrifying polyphosphate

摘要: The short-term effect of anaerobic reaction time (AnRT) (i.e., 90, 120 and 150 min) on the denitrifying phosphorus (P) removal performance and N O production was examined using a denitrifying enhanced biologic phosphorus removal (EBPR) sludge acclimatized with mixed acetate (HAc) and propionate (Pro) (in the molar ratio 3∶1) as carbon sources. The results showed that when the AnRT was prolonged from 90 to 150 min, the anaerobic polyhydroxyalkanoate (PHA) synthesis was decreased by 15.3%. Moreover, the ineffective PHA consumption occurred in anaerobic phases and contributed to an increased accumulation and higher free nitrous acid (FNA) concentrations (≥0.001–0.0011 mg HNO -N/L) in the subsequent anoxic phases, causing a severe inhibition on anoxic P-uptake and denitrification. Accordingly, the total nitrogen (TN) and total phosphorus (TP) removal efficiencies dropped by approximately 6.3% and 85.5%, respectively; and the ratio of anoxic N O-N production to TN removal increased by approximately 3.8%. The fluorescence in situ hybridization (FISH) analysis revealed that the sludge was mainly dominated by (62.0% (SE = 1.5%)). In conclusion, the short-term excessive anaerobic reaction time negatively impacted denitrifying P removal performance and stimulated more N O production, and its effect on P removal was more obvious than that on nitrogen removal.

Effect of wastewater composition on the calcium carbonate precipitation in upflow anaerobic sludge blanket

摘要: Calcium carbonate often precipitates in anaerobic reactors treating wastewater with high calcium content. The aim of this paper is to study the effect of wastewater composition on calcium carbonate precipitation in upflow anaerobic sludge blanket (UASB) reactors. Two laboratory-scale UASB reactors were operated with calcium-containing influents using acetate and carbohydrate as substrate, respectively. There was an obvious accumulation of inorganic precipitate observed in the biogranules. Observations via scanning electron microscope (SEM) and energy dispersive spectroscopy (EDS) showed that the acclimated biogranules in the two reactors differed in microstructure. Calcium carbonate was found to have precipitated on the surface of acetate-degrading biogranules, but precipitated at the core of the carbohydrate-degrading biogranules. The results indicated that substrates had significant influence on the location of calcium carbonate precipitation in anaerobic granular sludge, which was expected due to the different methanogens distribution and pH gradient within the granular sludge degrading various substrates. Moreover, the location of calcium carbonate precipitation substantially affected the specific methanogenic activity (SMA) of the granular sludge. The SMA of the acetate-degrading biogranules dropped from 1.96 gCOD·gVSS·d to 0.61 gCOD·gVSS·d after 180-d of operation in the reactor. However, the SMA of the carbohydrate-degrading biogranules was not adversely affected by calcium carbonate precipitation.

Influencing mechanism of high solid concentration on anaerobic mono-digestion of sewage sludge without

摘要: High-solids anaerobic digestion of sewage sludge was a promising process, but high solid concentration negatively influenced methane production. The influencing mechanism was systematically analyzed in this study through a series of static anaerobic digestion experiments at total solids (TS) contents of 3%–15%. The results showed that TS 6% was the boundary between low-solids and high-solids anaerobic digestion, and the accumulative methane yield decreased exponentially when TS increased from 6% to 15%. The performance of anaerobic digestion was directly determined by the efficiency of mass transfer, and the relation between methane yield and sludge diffusive coefficients was well described by a power function. Thus, the increasing TS resulted in an exponential increase in sludge viscosity but an exponential decrease in diffusive coefficient. The blocked mass transfer led to the accumulation of volatile fatty acids (VFAs) and free ammonia. Acetic metabolism was the main process, whereas butyric and propionic metabolisms occurred at the initial stage of high-solids anaerobic digestion. The concentration of VFAs reached the maximum at the initial stage, which were still lower than the threshold influencing methanogens. The concentration of free ammonia increased gradually, and the methanogenesis was inhibited when free ammonia nitrogen exceeded 50 mg·L . Consequently, the deterioration of high-solids anaerobic digestion was related to the blocked mass transfer and the resulting ammonia accumulation.
